What I want to see in Show and what I want to see in fic are quite different.
In fic, whump with consequences is my . . . kink? I guess. And because I'll never really adequately have that in Show, I don't expect it or wish desperately for it there. Not H/C, mind you, because the emphasis is always on the comfort that usually ends up being sexual in nature, or fluffy.
But In Show, yeah, prank wars and geekiness.
And competence in both Show and fic.
What a silly question. I also want to see more brothers being brothers.
I want to go on record as saying I do not need to see Dean (or anyone) puking on the show, or in fic, for that matter. That's ... really weird.

I wasn't actually thinking of them separately. I guess I was assuming that lame!Dean and puke!Dean would also be welcomed on TV, which is unsupported.
I don't know if I have any different wishes other than--gimme all the D/C fic in the world, but it's probably quite safest if that never happens onscreen. However, I read so much fic that doesn't meet my criteria in any other way that I might consider myself irretrievably tainted. I can't tell what distinguishes between a fic I finish and one I don't, because I have been reading some horrible shit recently, but still not *all* the D/C.

Show and fic serve different needs for me. I actually am less likely to read a fic if it tries to adhere too closely to canon or rewrite it. Because, man, Show is dark and depressing, and I don't generally want epic fics that try to tackle a given seasons Big Bad. But I do love case fics (MOTW style) (because that means a good whodunit to wrap the character stuff around). It goes back to my love of AUs, where fic doesn't mess too much with Show canon and mess with my head.

When I first started binging, before my ship needs crystallised, I was plowing through...K Hanna Korossy? Something like that. And she was straight up gen casefiles, with added cuddling. It was so weird. And her monsters were totally ho hum, but they'd beat the shit out of Sam and Dean, and then they'd need to patch each other up and share a bed! Again!

I don't mind canon and fic intersecting, although I can only remotely tolerate S/G in AU. And there are some canon things I hate being experimented with (birth order or age gap, most commonly, and then after that--randomly not killing people who die in canon, and either of the boys being angels (unless it's a boys-are-angels angels-are-hunters straight swap)), so maybe I'm less exploratory than many?

I'd actually like to see more "what if" stuff as is breaks off from canon. Like, what happens if Dean doesn't make the deal for Sam? What happens if Dean does die and Sam can't save him (which I wrote, actually)? What if John had died in Sam's nursery instead of Mary? Just to twist it up a little.
