RTD (and I know he's bowing out, but still) is on record as saying he thinks it would be a mistake to make the Doctor female. Actually, I tend to agree with him on that. Rachel Weisz as Romana though, no question.
And, following on from the Romana discussion, is it canon that male TimeLords regenerate as male, and female as female...?
In the past, when the BBC were casting a new Doctor, they would sometimes drop hints that they were considering a woman for the role. But it was mostly a way of generating publicity; they were never really serious about it.
I'm not ready for a new Doctor quite yet, but probably will be after one more season.

That would be fabulous. Now you've got my brain spinning off onto my wish-list for Romana casting.
As for the Eleventh Doctor, I would love to see any of the following: Idris Elba, Damian Lewis, Alexander Siddig, Rufus Sewell (Edit: Tilda Swinton would be fabulous as well)
As long as we're casting fantasy Doctors, how about Callum Keith Rennie? Who can play charming and quirky as easily as serious or haunted?
I feel like if they were going to make the Doctor female, they should have done it with Nine, right at the start of rebooting the franchise. Doing it with Eleven (or Twelve) would feel very much like a stunt.

My vote for the Eleventh Doctor would be Chiwetel Ejiofor. Playing Lola from
Kinky Boots.
I keep hearing about Robert Carlyle rumour. Carlyle is a fine actor, but I think he's too much of a David Tennant type -- small and intense and wiry. I'd welcome some out-there choices. An older Doctor maybe? (Someone on my Flist mentioned Bill Nighy, and I think he'd be a riot.) A female Doctor or an African-Brit would be awesome, but I can't really see BBC going there.
Bwah! I did offer him as a candidate in another discussion, but figured on a more conservative wardrobe. (He'd look great in a gray suit and bowtie, wouldn't he?)

That's the only way it's happened on the show as opposed to spoofs. But we've seen Romana briefly regenerate into an obviously unearthly humanoid, and Nine's discussion of the topic implied that completely inhuman regenerations were possible. If anything prevents genderswitch, I'd imagine it's the self-image of the Time Lord in question rather than any limitation of the process itself.
