Well, Henry said that she had killed two of "us." (I believe) which is where the confusion about the second person comes in.

When the Tailies were first attacked and Eko killed an Other, did A-L kill one then, too?

According to wikipedia, Ana Lucia killed an Other during a raid on the Tailies camp. She took a US Army knife off of the body:
After the tail section of the plane crashes into the water off the beach, the survivors swim ashore. *** That night, three of the adults are taken and Eko kills two of the "Others" with a rock when they try to take him. From that night on, he refuses to speak.
A survivor who identifies himself as Nathan suggests staying on the beach, and the group does so. On the fifth day, Donald dies from the leg injury and is buried. On the twelfth day, the Others take nine more, including the two children. Ana manages to kill another one, who is discovered to be carrying an antique U.S. Army knife and a list of the nine to be taken, along with their descriptions. While trying to sort out what happened, the notion of an infiltrator is discussed, as well as a proposal to leave the beach.
So, she killed 2 Others -- Goodwin and RedShirt Other #1

*If* Claire is his half-sister (and the way everything is connected, it's entirely plausible that she is), Claire could make a comment about how she never knew who her father was, but her mother always referred to him as a drunk surgeon, or something.
