I just saw a series of Buffys. Giles is left handed. He hands people things with his left hand, in Hush he does the projector stuff left handed, etc. It is very casual, and I bet if the scene calls for him to do stuff with his right hand, he can. Should think it would depend on the scene blocking.

Giles is totally left-handed.
My brother is left-handed, and he developed a habit of noticing left-handed people on screen. He'd just mutter to himself "that person is a leftie" whenever he noticed somebody like that, and I picked up that habit from him. You can only imagine how watching the sword-fight scene in "The Princess Bride" was for us.
Anyway, neither of us failed to notice that Giles is left-handed. Which hand does he use to take his glasses off all the time? Left. And I'm 99% sure this is the hand he uses when killing Ben in "The Gift".

I'd meant to ask about that; I forget which one he uses but "The Gift" will be on soon on Canada's Space Channel. I'll conduct some research.
Incidentally that scene shocked the hell out of me when I first saw it.
Nope, I was wrong - right hand. He pulled an Inigo Montoya on him and started their fight with the weaker hand, so that if he had lost with that hand, he'd be able to switch! That's my theory and I'm sticking to it.
He definitely is left handed, though.
I dunno, that close-up of the killing of Ben may be a reversed print. Nilly, can you find any other screen caps that show on which wrist Giles wore his watch?
eta: changed my mind about the reversed print. That's Giles's left hand. You can see his thumb pinching Ben's nose.
I'm not sure what you mean by reversed print, but on the TV screen, Giles suffocated Ben with his right hand. If course, that probably was a matter of blocking. Ben's head was to the right of the screen (Giles' left).
