I think the key is making a small movie in the universe that isn't necessarily any story from the comic. Which is basically the key to making ANY good comic book movie.
I would see del Toro or Cuaron do it.
We're fantasy casting over on my brother's FB wall.
So far my favorite picks are SWINTON for Desire (of course), Luna Lovegood as Delerium (I think I stole that from Hec?), and Tom Mison as Dream.
Chris Hemsworth was suggested for Destruction, but that's a bit on the nose. Likewise Patrick Stewart as Destiny.

Death is really hard to cast! Jennifer Lawrence could do a decent job, but she's EVERYWHERE these days, so I don't know.
Despair is the other tricky one. I'm drawing a complete blank for her.
Okay - in my fancast, Rinko Kikuchi [link] as Death. She can do the whimsical, the compassionate, and the "don't fuck with me" all at once.
Despair - if Rebel Wilson can actually do ennui, she could be great. She's got a great off-kilter delivery as it is.
