Actually I can see a way for a season six if it really happens. Season 5: resolve the apocalypse plot. Season 6 - back to monster of the week. No overarching plot. Brothers Winchester deal with fact that they will be fighting evil sons of bitches until some evil son of a bitch gets lucky and kills them. No retirement, no resolution. Save some people; get there a second too late for some people. Keeping fight the good fight until the day they die screaming.

That's sort of a huge problem. Every story needs conflict to keep people watching or reading, and facing an external beastie every week really ain't enough.
It would be like reading a book about an entirely happy family with no problems whatsoever. Boring as hell.
