Totally of the top of my head, if you ask me in 5 minutes I'll have different answers.
Dean - the LOOK on his face in "Dead Man's Blood", as he comes crashing through the brush with the knife and John & Sam are down.
Sam - his hitching breath as he watches John & Dean hug, how he wants to hug, but he's scared of getting smacked down by Dad, and... guh.
John - again, DMB. Him telling Sam about the college fund & how he spent it on ammo.
Hmmmm. I think I need to re-watch the end of S1.

I became very fond of the "saving people, hunting things...the family business" that's in every previously.
The way Dean just dives for the other chair so Sam has to sit in the clown chair in Everybody Loves A Clown.

Oh, that made me laugh SO MUCH.
I watched Route 666 not too long after y'all last mentioned it., because I realized I hadn't actually seen it. It reinforced my growing belief that my standards are pretty low, because I was all just "racist ghost truck, of course, la la la la la, makes perfect sense" even after being warned of the eye-rolliness.
I feel I have to mention, I just figured out that WAH is a TLA and not a sound-effect.
